Apomorphine-induced Rotation Test
behavioralRatSprague-Dawley (SD)
Objective: To assess rotational behavior in rats with 6-OHDA-induced Parkinson's disease model using apomorphine-induced rotation as a measure of dopaminergic denervation
Protocol Steps
1
Establish PD model with 6-OHDA injection
Inject 8 µL of 6-OHDA solution (2 µg/µL with 0.01% ascorbic acid) into the right middle forebrain bundle using specific coordinates
View evidence from paper
“Each rat was injected with 8 µL of this solution into the right middle forebrain bundle (MFB)”
2
Wait for lesion development
Allow three weeks following the development of 6-OHDA lesions before testing
View evidence from paper
“over the three weeks following the development of 6‐OHDA lesions”
3
Administer apomorphine
Give subcutaneous injection of apomorphine at 0.25 mg/kg dose
View evidence from paper
“Apomorphine (0.25 mg/kg, APO; catalog no. 2073/50, Tocris) was administered subcutaneously to the rats”
4
Count rotations
Count the number of contralateral 360° rotations within a 30-minute period
View evidence from paper
“we counted the number of contralateral 360° rotations within a 30‐min period”
5
Apply inclusion criteria
Include only rats that completed more than 210 total rotations for further analysis
View evidence from paper
“For further analysis, only rats that completed more than 210 total rotations were included”
