Protocol Steps
Complete CPA extinction
Ensure extinction criteria are met through daily 20-25 day sessions until paired t-test confirms extinction
View evidence from paper
“Extinction sessions began 24 h after the CPA post-test and were conducted daily for 20–25 days until extinction criteria were met”
Administer propranolol (if applicable)
Inject propranolol (10 mg/kg, i.p.) or vehicle one day after extinction, timing varies by stressor type
View evidence from paper
“mice were randomly assigned to receive either the β-ARs antagonist propranolol at a dose of 10 mg/kg (i.p.) or its vehicle (saline)”
Apply stressor
Expose mice to social defeat (15-min agonistic encounter), restraint stress (15 min in conical tubes), or tail-pinch (15 min with paper clip)
View evidence from paper
“exposing mice to either an acute social defeat or a physical stressor (restraint or tail-pinch)”
Conduct reinstatement test
Place mice in central passageway and allow free access to all chambers for 15 minutes in a different room from conditioning
View evidence from paper
“allowing animals free access to all compartments of the apparatus for 15 min”
Record behavioral measures
Measure time spent in naloxone-paired chamber and number of crossings between compartments
View evidence from paper
“Time spent in the naloxone-paired chamber and the number of crossings between compartments were recorded”
