Novel Object Recognition Test
Objective: To examine recognition memory using a Novel Object Recognition test
Protocol Steps
Test timing
Conduct the Novel Object Recognition test four days after conclusion of sleep disruption
View evidence from paper
“Four days after the conclusion of the SD, mice were subjected to a Novel Object Recognition (NOR) test”
Room habituation
Habituate mice to the testing room for 30 minutes prior to experimentation
View evidence from paper
“Mice were habituated to the testing room for 30 min prior to experimentation”
Arena habituation
Place mice individually in empty PhenoTypers to habituate to the arena for 10 minutes
View evidence from paper
“mice were individually placed in empty PhenoTypers to habituate to the arena for 10 min”
Break period
Provide a 1-hour break after arena habituation
View evidence from paper
“After a 1‐h break, the familiarization phase was conducted”
Familiarization phase
Place mice in the same PhenoTyper containing two identical objects (either LEGO towers or T25 flasks) and allow free exploration for 10 minutes
View evidence from paper
“mice were placed in the same PhenoTyper containing either two LEGO towers or two T25 cell culture flasks filled with bedding and allowed to freely explore the objects for 10 min”
Second break period
Provide another 1-hour break after the familiarization phase
View evidence from paper
“After another 1‐h break, the NOR test was conducted”
Novel Object Recognition test
Conduct the NOR test with one familiar object swapped for a novel object and allow exploration for 10 minutes
View evidence from paper
“the NOR test was conducted with one object being swapped out for a novel one (a LEGO tower or T25 flask) and mice being allowed to explore the two objects for 10 min”
Recording and analysis
Record all trials using EthoVision XT software and score manually
View evidence from paper
“All trials were recorded using EthoVision XT software and scored manually”
